To get the best picture on an HDTV, you want to first use the best video cable possible. HDMI will provide you the best quality. If that is not available, then component video cables.

Next, watch HD sources, such as HD cable, 720p Xbox or 720p BluRay DVD player. The less scaling the TV has to do, the better the image will be. Scaling occurs, when the video resoution doesn't match the native resolution of the TV.

If you want to get really fancy, you can buy a home theater calibration DVD or color calibrator to ensure that you have the best possible color as possible. Personally, I rather let my eyes be the judge.
The cheapest was: Use the THX video/audio testing program found on any THX certified DVD. I think the Star Wars and Indiana Jones DVDs has the program.

It works well and it's the best picture you'll get w/o hiring a professional calibration expert.

For the most resolution, you'd just have to use an HD source, either 1080p (Blu-Ray) for 1080p capable sets, or 720p (Any HD) for 720p capable sets. For the clearest picture, try not to use sharpness over 50% on ANY TV, as it will introduce artifacts. Always turn off anything w/ Edge Enhancement, or anything that has to do with sharpness.
